The transition from water to land environments enabled the evolution of locomotor strategies in vertebrates. Understanding the mechanisms underlying skeletal muscle development, or myogenesis, can provide cues about how skeletal muscle evolved. In amniotes, all skeletal muscles of the body derive from the dermomyotome, a reservoir of muscle stem cells (MuSCs). Trunk and limb muscles are evolutionary distinct being formed through different paths.  Dermomyotomal MuSCs delaminate and form the ancestral embryonic myotomes in the trunk, defining the epaxial muscles or migrate directly towards the limb bud, independently of the myotome. This led us to ask to what extent the myotome is necessary for the formation of the epaxial muscles?
